Ensitrelvir now available in US for COVID-19 postexposure prophylaxis

The SCORPIO-PEP Phase 3 clinical trial demonstrated that ensitrelvir, a SARS-CoV-2 main protease inhibitor, significantly reduced the risk of symptomatic COVID-19 following household exposure. In a study involving 2041 participants, ensitrelvir achieved a 67% relative reduction in symptomatic cases compared to placebo over a 10-day period. The trial utilized a once-daily regimen administered within 72 hours of exposure and showed an overall efficacy rate of 98% against SARS-CoV-2 antibodies. Common adverse events associated with ensitrelvir included headache, diarrhea, and cough, alongside potential serious risks such as allergic reactions.
